Culture of Euglena with digestate from Methane fermentation

The effects of light intensities and digestate concentrations on growth of Euglena gracilis were investigated to select the suitable condition for culturing with digestate from methane fermentation. E. gracilis was cultured in an aqueous solution containing digestate at different concentrations of 5–100%, and Cramer–Myers (CM) solution as a control at photosynthetic photon flux densities (PPFDs) of 75–150 µmol m-2 s-1 with continuous illumination at 30°C. The number of cells was monitored daily and specific growth rates were calculated. The specific growth rates were greater in appropriate concentrations of digestate than in CM solution and the maximum specific growth rate was 0.047 h-1 in 10% digestate at a PPFD of 150 µmol m-2 s-1. These results demonstrate that E. gracilis can be cultured at high growth rates with diluted methane fermentation digestate under a relatively low PPFD level. In simulation experiment conducted with E. gracilis density of 30×105 cells ml-1, more than 10% of light was transmitted at the depths shallower than 15 mm, using 20% diluted digestate.
